Well, the reason I am contributing on this one is that this does not seem 100% clear to me, particularly as I found references to trials "con placebo" and "contra placebo". On the face of it, this doesn't mean the same thing to me.

Discussion
The key word here is "control". A placebo-controlled study is one where you have a "control group" that is given a placebo instead of the trial drug. The behaviour of this group then provides a standard or baseline (called the control or control-variable) against (or with) which the effectiveness or otherwise of the trial drug can be compared.
An example of a scientific control (sometimes called an "experimental control") might be testing plant fertilizer by giving it to only half the plants in a garden: the plants that receive no fertilizer are the control group, because they establish the baseline level of growth that the fertilizer-treated plants will be compared against. Without a control group, the experiment cannot determine whether the fertilizer-treated plants grow more than they would have if untreated.
