in-fill = sitios disponibles

En el caso se refiere a sitios todavía disponibles en zonas urbanas ya existentes desde hace algún tiempo, o sea terrenos baldíos no en las afueras sino en zonas más céntricas.

La idea es ¨rellenar¨ totalmente la ciudad.

no estoy seguro, por la falta de contexto, pero podrían ser rellenos sanitarios. En algunas ciudades han empezado a construir en lo que antes eran basureros. si este no es el caso, entonces podría usarse "lotes baldíos".
