Durch Totalreflexion des anregenden Laserstrahls an der Grenze zwischen Deckglas und angrenzendem waessrigem Medium wird die Fluoreszenzanregung auf eine dünne Grenzschicht von ca. 100 nm limitiert. Die daraus resultierende minimale Anregung von Fluorophoren ausserhalb der Objektebene, d.h. die minimale Hintergrundfluoreszenz erlaubt mit dieser Technik einzelne Fluorophore bzw. einzelne Moleküle verfolgen zu können.
