Cryo-ET was used to visualise the cytoskeleton reorganisation in the cells infected by Listeria.
Measurements have been performed at cryo-temperatures on cell samples preserved in a **close-to-life state.** The quantitative analysis of the supramolecular architecture revealed the existence of bundles of nearly parallel hexagonally packed filaments with spacing of 12 to 13 nm.

http://esgweb1.nts.jhu.edu/cellbio/devreotes/Breitschneider-...
Da cui è tratto quanto segue:
"The delicate balance of proteins controlling actin organization in Dictyostelium as in other fast moving cells, such as neutrophils, requires reliable preservation of the momentary functional state of actin networks subjected to electron microscopic analysis. Vitrification by rapid freezing ensures close-to-life pre- servation of actin structures under scrutiny, avoiding the risk of artifacts associated with detergent treat- ment, chemical fixation, staining or dehydration"
