So, in the movie "Fright Night", we see the hero, Charlie, go to a vampire expert for help. The vampire seems to be kind of a womanizer, as it was referred to in an earlier scene. As soon as the hero asks him for help, the vampire goes 'I'm gonna pop your cherry.

I believe it's said figuratively, but in what sense is the phrase?

The character who says "I'm gonna pop your cherry" isn't a vampire. It's the character called Peter Vincent who says it. For more info have a look at these links -

If this is the correct script then it may be a reference to Charlie or Ed (I can't tell which one it is from this script) telling Peter Vincent that this is his first assignment for a newspaper.

@ Asker Phil is right, that is the usual meaning.
Here, I'd imagine it means "lose his vampire virginity" — we often talk about someone being an "xxx virgin" if xxx is something they've never done before.

Have you Googled this? It means take your virginity. I don't know how literal this is meant to be, but he continues by saying "Ginger, take him upstairs, you've got ten minutes". I haven't been able to find the wider context, so perhaps you could tell us what's going on.
