This vector was used as a backbone, and the viral vectors (pUC19 derivative vectors: CaLCuV A, CaLCuV A plus FLC1, CaLCuV plus FLC5, CaLCuV plus FLC-Consensus) containing the FLC sequences were digested with PvuII, the fragments purified and cloned into the PHP13184 vector.

disagree  Arkadii Marchenko: сшивают ДНК-лигазой, а сшить полимеразой Кленова если и можно (о чем я не слышал) то очень сложно

  -> У ДНК-полимеразы много разных активностей (функций), но фрагмент Кленова как раз некоторых из них лишен, он способен лишь достраивать комплементарную цепь. По-видимому, Вы правы, и речь здесь идет лишь о достройке тупых концов фрагментов, а не о сшивании.
