Supernatants from recombinant strains were separated on NuPage BisTris 10% SDS-PAGE gel and stained with Coomassie (FIG. 1). Due to the glycosylation by the host strain the phytase occurred as three bands between 44.2 and 53.2 kDa. All samples were applied with equal phytase activities. The gels were dried and scanned on an Agfa flat bed scanner with the Phoretix ID Advanced software. The areas of the three bands were integrated and the data is summarised in the following table 3. For all four tested strains the sum of the area of all three phytase bands in equal within the scope of the measurement accuracy. This shows that the mutation in Da2 does not result in alterations in the specific activity of the enzyme and thus demonstrates that the mutation is responsible for the increased secretion of the phytase protein.

Überstände aus rekombinanten Stämmen wurden auf NuPage BisTris 10% SDS-PAGE-Gel getrennt und mit
Coomassie gefärbt (Fig. 1). In Folge der Glykosilierung durch den Wirtsstamm erschien die Phytase als drei Banden
zwischen 44,2 und 53,2 kDa. Alle Proben wurden mit gleichen Phytaseaktivitäten aufgetragen. Die Gele wurden getrocknet und mit der Phoretix ID Advanced Software auf einem Agfa Flachbettscanner gescannt. Die Flächen der drei Banden wurden integriert und die Daten sind in der nachstehenden Tabelle 3 zusammengefasst. Die Summe der Flächen aller drei Phytase-Banden ist für alle vier getesteten Stämme im Rahmen der Messgenauigkeit der Methode gleich. Dies zeigt, dass die Mutation in Da2 keine Veränderungen in der spezifischen Aktivität des Enzyms verursacht und belegt somit, dass die Mutation für die erhöhte Sekretion des Phytaseproteins verantwortlich ist.
