Introduction
Recent successes with drugs targeting T-cell inhibitory molecules has led to a significant increase in enthusiasm for immunotherapy. Broadly speaking, immunotherapy is a treatment designed to enhance a patient’s immune response against their tumors. There are multiple forms of immunotherapy, which include: 1) checkpoint blockade, the class of drugs targeting T-cell inhibitory molecules; 2) adoptive cellular therapy, which involves direct infusion of T cells into patients; and 3) vaccines, which are a form of active immune therapy designed to stimulate the patient’s immune system to recognize tumor cells as foreign. The majority of cancer vaccines include an antigen that serves as the target for the immune response, as well as an immunoadjuvant, which is a nonspecific stimulator of the immune response that promotes an environment conducive to immune stimulation. The appeal of vaccines as a therapeutic strategy is that they are target-specific and associated with minimal toxicity. In addition, they stimulate the adaptive immune system, thereby producing a memory response allowing for sustained effect without repeated therapy. For these reasons, physicians and patients alike are enthusiastic about the possibility of a breast cancer vaccine that could stimulate the patient’s immune system to recognize, attack, and eliminate tumor cells. Although there are currently no US Food and Drug Administration (FDA)-approved breast cancer vaccines, there are multiple vaccines and vaccine treatment strategies that are being actively investigated in clinical trials.
NeuVax – Vaccination for Secondary Prevention
The breast cancer vaccine farthest along in development is NeuVax, which is comprised of the human epidermal growth factor receptor 2 (HER2)-derived peptide E75 (nelipepimut-S) combined with granulocyte-macrophage colony-stimulating factor (GM-CSF) as an immunoadjuvant. E75 is a major histocompatibility complex (MHC) class I epitope that stimulates a CD8+ cytotoxic T-lymphocyte (CTL) response.

Source text - English It is not enough to diagnose prostate cancer early, if the cure is perceived as worse than the disease itself. This was the case 20 years ago, when the radical prostatectomy (developed by Hugh Hampton Young at Johns Hopkins nearly a century ago) was infamous among surgeons and patients for its extreme, often life-threatening bleeding and devastating side effects: One-quarter of the men who survived the operation were left with severe incontinence, and all of them were impotent. The very harshness of the procedure was the catalyst- for change, inspiring the anatomical discoveries that have drastically reduced these side effects.
Today, radical prostatectomy cures the vast majority of men with cancer confined to the prostate preserving potency. - by not removing one or both of the nerve bundles adjacent to the prostate (surgeons didn't even realize these bundles existed two decades ago), which are responsible for erection - does not compromise cancer cure; a recent study found that the odds of cure are just as high. Today, serious bleeding is very rare, preserving potency is common, and few suffer from serious incontinence. In fact, the radical prostatectomy series at the Brady Urological Institute is considered to be the gold standard for cancer cure, to which all other forms of treatment are compared.
However, we are far from complacent: Our surgeons, like our basic scientists, are always raising their standards of excellence. Every day, we aim higher, because there is still so much more that we want to do to help our patients. For example, we are constantly seeking new ways to improve our surgical technique and reduce side effects - through the use of nerve stimulators that can help pinpoint these microscopic nerves more accurately; with techniques that may enable the nerves to regenerate more quickly; and with alternate medical therapies to help maximize the ability of damaged nerves.

These cab stands are located directly in front of the arrival halls at Terminal 1 – exit D, and at Terminal 2 – exit E. The company provides a taxi service at guaranteed tariff prices which are equal to the maximum prices allowed for taxi services in Prague. Fees start at 40 CZK plus 28 CZK per kilometre, plus 6 CZK per minute of waiting or traffic, based on the number of kilometres travelled. Estimated price from airport to city centre or vice versa is around 650 CZK to 700 CZK.
You can easily recognise them as they are all white with the FIX TAXI logo, and they use Czech-made cars – Skoda Superb or VW Transporter minivans. It is also important to mention that FIX TAXI was formerly known as Airport Cars or 111-Radiocab, a company that had a bad reputation in the press for reckless driving in the city centre using silver VW Passat cars. The company started business in 1991, not long after the Czech Republic Velvet Revolution, when overcharging was at its peak and it was unusual to charge customers 2000 CZK for the ride to the city centre.
Drivers are required to speak at least basic English and all vehicles provide free Wi-Fi connection. The company no longer offers discounts for return taxi bookings.

Among the native Hopi tribes of the American Southwest, an ancient ritual of holy dancers and merry clowns is still performed today. The Hopi way of life might seem very foreign to most of us, but the comic antics of its tribal clowns are remarkably familiar. The dance of the "kachinas" (nature spirits) is held in the plaza formed by the pueblos of the Hopi village, and lasts a full day, if not two. Early in the afternoon of the first day, as the masked kachinas dance to musical accompaniment, the audience at this sacred ceremony is suddenly distracted by the noisy and somewhat supernatural appearance of several Chükü'wimkya clowns on the rooftop of one of the buildings that line the plaza. The bodies of these clowns are smeared with mud from the sacred springs, while on their heads they wear improvised wigs made from stocking caps and rabbit fur. They accentuate their facial expressions with a black inverted v over each eye and u-shaped black marks under the eyes and mouth.
The clowns pretend to step off the edge of the roof, one foot suspended in mid-air, then retreat in mock fear, provoking uproarious laughter from the spectators below, who quickly lose interest in the dancers. The clowns lower a long plank to the ground and attempt to slide down it headfirst, and considerable comic horseplay follows in which they almost lose their balance before finally tumbling to the ground. Parading around the plaza, the clowns suddenly feign great surprise at seeing the kachinas dancing, and immediately decide to join in.

DA ESTRUTURA DE PNEUS
1. Introdução
O Pneu é um insumo de grande representatividade no processo produtivo de uma mina. De acordo com a Caterpillar (2013) os gastos com pneus dos caminhões fora-de-via representam 16% do seu custo operacional total.
Dependendo das solicitações, um pneu pode atingir o final de vida útil por motivos distintos. Alguns fabricantes de pneus os classificam em três diferentes grupos, sendo eles: desgaste da banda de rodagem; danos provocados por agressões externas representadas por cortes que inutilizam os pneus; e a fadiga representada como a exaustão da resistência mecânica da carcaça do pneu. Os dados apresentados na Figura 1 oferecem uma perspectiva do universo de pneus e seus respectivos motivos de sucateamento para uma mineração a céu aberto. Os modos de falha destacados como trincas e deslocamento têm relação direta com o processo de fadiga. Nota-se ainda um grande potencial de aumento da utilização destes componentes, pois foram sucateados com remanescente de mais de 40% em média da espessura da banda de rodagem.
Atualmente a demanda por remoção de um pneu que atingiu o limite de vida útil por fadiga é feita por inspeções visuais. Conforme Michelin (2012) profissionais devidamente treinados são capazes de determinar tecnicamente a continuidade de operação de um pneu. Esses técnicos vão a campo regularmente buscando identificar fisicamente características típicas de pneus fadigados. Como o procedimento atualmente utilizado para priorizar as inspeções é a aproximação do final de vida útil do pneu, uma orientação com informações de solicitações geradoras da fadiga podem ser úteis para apoiar a identificação dos pneus que se aproximam da quantidade limite de ciclos de carregamento. Esta medida pode ser antecipada ou postergada em detrimento da quantidade de ciclos e intensidade das solicitações sobre o pneu durante toda sua operação.
Como as falhas por fadiga ocorrem muitas vezes abruptamente e, como o pneu contem certa massa de ar confinada sob pressão, falhas repentinas podem resultar em grandes deslocamentos de massas de ar. Potenciais danos pessoais e materiais podem ser evitados caso ocorra uma atuação antes do atingimento do limite de vida útil por fadiga em um pneu.

1. Introduction
The tire is an input of great representativeness in a mine’s productive process. According to Caterpillar (2013), the expenses with off-highway trucks represent 16% of their total operational cost.
Depending on the requests set upon it, a tire might reach the end of its service life for distinct reasons. Some tire manufacturers classify them in three different groups, being: wear and tear of the tread; damage caused by external aggressions, represented by cuts that render the tires useless; and the fatigue represented as the exhaustion of the mechanical resistance of the tire’s carcass. The data presented in Picture 1 offer a perspective of the universe of tires and their respective scrapping reasons for an open pit mine. The highlighted types of failures, such as cracks and separations, are directly related to the fatigue process. It is also noticed a great potential for the increase in these components’ utilization, since they were scrapped, in average, with over 40% of the tread thickness remaining.
Currently, the demand for removal of a tire that has reached the end of its service life through fatigue failure mode is made through visual inspections. According to Michelin (2012), properly trained professionals are capable of technically defining the continuity of a tire’s operation. These technicians regularly go to the field in order to physically identify typical characteristics of fatigued tires. Since the procedure currently used to prioritize the inspections is the approximation of the end of the tire’s service life, measured in worked hours, other information about the fatigue process may be useful to support the identification of tires that are close to reaching the loading cycles’ limit. The tire replacement can be anticipated or postponed according to information about the amount of cycles and the intensity of the loads over the tire during its service life.
Since the failures by fatigue often happen abruptly, and since the tire contains an amount of air mass confined under pressure, sudden failures may result in great air mass dislocations. Potential personal and material damages may be avoided if something is done before the tire reaches the end of its service life through the fatigue process.

Publicado em fins de junho de 2013, no centro dos acontecimentos políticos que abalaram o Brasil naquele mês, o livro eletrônico Vinagre: uma antologia de poetas neobarracos, organizado por Fabiano Calixto e Pedro Tostes, ainda é, passados já dois anos, a expressão artística mais imediata e ao mesmo tempo mais significativa daquele contexto, surgindo como uma sua consequência direta, uma espécie de instantâneo tomado no calor da hora, pleno de espontaneidade e desorganização, nascido sem planejamento prévio, de modo algo semelhante, quem sabe, aos das próprias manifestações que inundaram as cidades brasileiras. Atuando como um elaborado arquivo dos afetos e das tensões, das violências, imagens e esperanças daqueles dias de fúria e expectativa, os poemas reunidos em Vinagre, sob essa perspectiva, vêm se somar – guardadas, é verdade, as muitas e importantes distinções existentes – aos milhares de vídeos e fotos pessoais feitos durante os protestos, cada um deles constituindo pequenos fragmentos de um evento que, pela sua natureza movente e coletiva, permanece não-reconstituível em sua totalidade. Os próprios organizadores do volume, e também os seus primeiros e raros comentadores, procuraram ressaltar a urgência e a rapidez do material, dado à luz, em sua primeira edição, num intervalo de pouquíssimos dias desde a eclosão da violência e dos protestos massivos. Por tudo isso, talvez, e também pela irregularidade de algumas das composições apresentadas no livro (parte delas peças mais simples de agitação política ou comoção individual diante das demandas do momento), Vinagre não tenha recebido atenção crítica à sua altura, lida tão somente como poesia de circunstância – documento político de alguma relevância, mas nada além disso. Contrariando esse diagnóstico, por outro lado, gostaria de propor, como hipótese de trabalho, outra leitura, observando de modo diverso a sequência de acontecimentos e eventos culturais que levou até o surgimento de Vinagre. Longe de ser fruto das circunstâncias ou de paixões fortuitas, o livro é expressão (quem sabe mesmo a consolidação) de uma tendência específica no cenário da poesia brasileira contemporânea.

1. A poetics of friction
Published in the end of June 2013, in the center of political events that shook Brazil throughout that month, the electronic book Vinagre: uma antologia de poetas neobarracos (Vinegar: an anthology of neo-donnybrook poets), organized by Fabiano Calixto and Pedro Tostes still is, after two years, the most immediate and at the same time most significant artistic expression from that context, arising as its direct consequence, a sort of snapshot taken in the heat of the moment, full of spontaneity and disorganization, born without previous planning, somehow similar, maybe, to the very manifestations that flooded the Brazilian cities. Acting as an elaborate archive of affections and tensions, of violence, images and hope from those days of fury and expectation, under that perspective the poems gathered in Vinagre come to add up – guarded, of course, the many important existing differences – to the thousands of personal videos and photos taken during the protests, each one constituting small fragments of an event which, for its moving and collective nature, remains non-reconstitutable in its totality. The very organizers of the volume, as its first and rare commentators, have sought to point out the work’s urgency and speed, being born, in its first edition, in an interval of very few days after the outbreak of violence and massive protests. For all this, maybe, and also because of the irregularity of some of the compositions presented in the book (part of them, simpler pieces of political agitation or individual commotion in view of the moment’s demands), the critics’ attention given to Vinagre hasn’t measured up, the book seen only as circumstantial poetry – a political document of some importance, but nothing beyond that. Countering this diagnosis, on the other hand, I would like to propose, as a work hypothesis, another reading, observing in a different way the sequence of happenings and cultural events that led to the making of Vinagre. Far from being product of circumstances or fortuitous passions, the book is the expression (or even the consolidation) of a specific trend in the scenery of Brazilian contemporary poetry.

Source text - Portuguese Para a enzima CYP2D6, podem ser identificados quatro fenótipos: O tipo mais frequente é o ‘METABOLIZADOR NORMAL’ (dois alelos normais). É para estes indivíduos que são determinadas e recomendadas as doses médias a serem usadas de cada medicamento. Medicamentos prescritos em doses médias farão, portanto, o efeito esperado no seu organismo.
O segundo tipo é o 'METABOLIZADOR INTERMEDIÁRIO', que apresenta variações das CYPs que metabolizam os medicamentos mais lentamente que os normais; eles são heterozigotos para um alelo deficiente ou carregam dois alelos que causam redução da atividade da enzima. Neste caso, doses médias dos medicamentos poderão fazer seu efeito terapêutico. Todavia com o tempo, por ser eliminado mais lentamente, o medicamento se acumulara no corpo e poderá causar efeitos colaterais. Nesses casos, pode-se fazer o controle das concentrações dos medicamentos no sangue (monitorização) para evitarmos os efeitos indesejáveis.
O terceiro tipo é o 'METABOLIZADOR LENTO', nos quais há deficiência funcional da enzima. Nestas pessoas, a inativação e eliminação do medicamento são tão lentas, que ele se acumula em seu organismo em concentrações muito acima das desejáveis, levando a ocorrência mais frequente de efeitos colaterais e reações de intolerância, mesmo com o uso de doses baixas ou médias dos medicamentos. Nestes casos, a monitorização do medicamento permite um ajuste para doses mais baixas, aproveitando ao máximo os efeitos terapêuticos, mas evitando-se os efeitos tóxicos e colaterais.
O quarto tipo é o 'METABOLIZADOR ULTRARRÁPIDO', que possui múltiplas cópias do gene. Nestas pessoas, o medicamento é inativado e eliminado tão rapidamente que, em doses normais, praticamente não tem tempo de exercer o seu efeito completo. São aquelas pessoas que contam ao médico que "o remédio é como água, não tem nem efeito terapêutico e nem efeitos colaterais". Nessa categoria incluem-se muitos pacientes que não melhoram com nenhum medicamento em doses convencionais. Eles eram chamados de "resistentes à terapia". Hoje sabemos que muitos destes "resistentes" são, na verdade, METABOLIZADORES ULTRARRÁPIDOS. Estes indivíduos podem se beneficiar do uso de doses maiores dos medicamentos. O ajuste da dose tem transformado muitos destes pacientes "resistentes" em bons "respondedores" ao tratamento personalizado.
Translation - English For the CYP2D6 enzyme, four phenotypes can be identified. The most frequent one is the “NORMAL METABOLIZER” (two normal alleles). It is for those individuals that the average dosage of each medication is determined and recommended. Hence, medications prescribed in average dosages will cause the expected effect in their organisms.
The second type is the “INTERMEDIATE METABOLIZER”, who presents variations in the CYPs that cause medications to be metabolized slower than normal. They are either heterozygote for a deficient allele, or carry two alleles that reduce the enzyme’s activity. In that case, average dosages of the medication might work their therapeutic effect; however, with time, since it’s being eliminated more slowly, the medication will build up inside the body and may cause side effects. In those cases, the medication concentration in the blood can be controlled (monitoring) to avoid the unwanted effects.
The third type is the “SLOW METABOLIZER”, in whom there is a functional deficiency of the enzyme. In those people, the inactivation and elimination of the medication is so slow that it builds up in their organisms in concentrations way above the desirable ones, leading to a more frequent occurrence of side effects and intolerance reactions, even when the medication is used in low or average dosages. In those cases, monitoring the medication allows an adjustment to lower dosages, taking the most advantage of the therapeutic effects, but avoiding the toxic side effects.
The fourth type is the “ULTRA-FAST METABOLIZER”, who possesses multiple copies of the gene. In those people, the medication is inactivated and eliminated so quickly that, for normal dosages, it practically doesn’t have enough time to carry out its full effect. These are the people who tell the physician that “the medicine is like water, it doesn’t have any therapeutic effect or side effects.” This category includes many patients who don’t improve with any medication in its conventional dosages. They were called “resistant to therapy.” Today, we know that many of these “resistant” patients are actually ULTRA-FAST METABOLIZERS. These individuals may benefit from the use of higher medication dosages. The dosage adjustment has turned many of these “resistant” patients into good “responsive” ones when treatment is personalized.
