La vincamine contient au minimum 98,5 pour cent et au maximum l’équivalent de 101,5 pour cent de éthyl-13a hydroxy-12 octahydro-2,3,5,6,12,13,13a,13b 1H-indolo[3,2,1-de]pyrido[3,2,1-ij][naphtyridine-1,5]carboxylate-12-(12S,13aS,13bS) de méthyle, calculé par rapport à la substance desséchée.
CARACTÈRES
Poudre cristalline or microcristalline blanche, inodore, pratiquement insoluble dans l’eau, soluble dans le chloroforme, peu soluble dans l’éther and sans l’alcool.
IDENTIFICATION
A. Dissolvez 50,0 mg de vincamine dans l’acide chlorhydrique 0,1N et complétez à 500,0 ml avec le même acide. Prélevez 10,0 ml de cette solution et complétez à 100,0 ml avec l’acide chlorhydrique 0,1N. Examinée de 210 nm à 320 nm (V.6.19), la solution diluée présente deux maximums d’absorption à 220 nm et à 268 nm et un minimum d’absorption à 243 nm. L’absorbance spécifique au maximum à 268 nm est de 231 à 255.
B. Examinez la vincamine par spectrophotométrie d’absorption dans l’infrarouge (V.6.18). Les maximums d’absorption du spectre obtenu avec la substance à examiner correspondent en position et en intensité relative à ceux du spectre obtenu avec la vincamine SCR fr.
C. Examinez les chromatogrammes obtenus dans l’essai « Substances apparentées ». La tache principale du chromatogramme obtenu avec la solution à examiner est semblable quant à sa position, sa coloration et ses dimensions à la tache principale du chromatogramme obtenu avec la solution témoin (a).
D. La vincamine donne la réaction des alcaloïdes (V.3.1.1).
ESSAI
Pouvoir rotatoire spécifique (V.6.6). Dissolvez 0,200g de vincamine dans la pyridine R et complétez à 20,0 ml avec le même solvant. Calculé par rapport à la substance desséchée, le pouvoir rotatoire spécifique est de + 39º à + 43º5.
Substance apparentées. Effectuez l’essai à l’ abri de la lumière et préparer extemporanément la solution à examiner et les solutions témoins.
Opérez par chromatographie sur couche mince (V.6.20.2) en utilisant une plaque recouverte de gel de silice HF254 R.
Solution à examiner. Dissolvez 50 mg de vincamine dans du chloroforme R et complétez à 5 ml avec le même solvant.
Solution temoin (a). Dissolvez 0,1g de vincamine SCR fr dans le chloroforme R et complétez à 10 ml avec le même solvant.
Solution temoin (b). Dissolvez 20 mg de vincamine SCR fr dans le chloroforme R et complétez à 100 ml avec le même solvant.
Solution temoin (c). Prélevez 5ml de solution temoin (b) et complétez à 20 ml avec du chloroforme R.
Déposez séparément sur la plaque 10 μl de chaque solution.
Développez sur un parcours de 12 cm, avec un mélange de 90 volumes chloroforme R et de 10 volumes de méthanol R. Laissez sécher la plaque à l’air. Examiner en lumière ultraviolette à 254 nm puis pulvérisez du réactif à l’iodoplatinate R.
Epivincamine. S’il apparaît une tache correspondant à l’épivincamine dans le chromatogramme obtenu avec la solution à examiner, elle n’est pas plus intense que la tache principale du chromatogramme obtenu avec la solution témoin (b).
Autres substances. S’il apparaît d’autres taches que la tache principale et que la tache correspondant à l’épivincamine dans le chromatogramme obtenu avec la solution à examiner, aucune d’entre elles n’est plus intense que la tache principale du chromatogramme obtenue avec la solution témoin (c).
Perte à la dessiccation (V.6.22). Déterminée à l’étuve à 100-105 ºC sur 1,00g de vincamine, la perte à la dessiccation n’est pas supérieure à 0,5 pour cent.
Cendres sulfuriques (V.3.2.14). Déterminé sur 0,5g de vincamine, le taux des cendres sulfuriques n’est pas supérieur à 0.2 pour cent.
DOSAGE
Dissolvez 0.300g de vincamine dans un mélange de 25 ml d’acide acétique anhydre R et de 5 ml d’anhydride acétique R.
Effectuez le dosage des bases en milieu non aqueux (V.3.5.5) en titrant par l’acide perchlorique 0,1N en présence de 0,2 ml de solution de violet cristallisé R jusqu’à virage au vert franc.
Effectuez un titrage à blanc. 1ml d’acide perchlorique 0,1 n correspond à 33,44 mg de C12H26N2O3.
CONSERVATION
En récipient bien fermé, à l’abri de la lumière.

CHARACTERISTICS
White crystalline or microcrystalline powder, odourless, practically insoluble in water, soluble in chloroform, less soluble in ether and in alcohol.
IDENTIFICATION
A. Dissolve 50.0 mg vincamine in 0.1N hydrochloric acid and dilute it to 500.0 ml with the same acid. Take 10.0 ml of this solution and dilute it to 100.0 ml with 0.1N hydrochloric acid. C. Examine the chromatograms obtained in the tests “Related substances”. The principal spot in the chromatogram obtained with the solution to be examined is similar in its position, its staining and its size to the principal spot in the chromatogram obtained with the reference solution (a).
D. Vincamine gives the reaction of alkaloids (V.3.1.1).
TESTS
Specific optical rotation (V.6.6). Dissolve 0.200 gm vincamine in pyridine R and dilute to 20.0 ml with the same solvent. The specific optical rotation, which is calculated in relation to the dried up substance, is from +390 to +43.50.
Related substances. Carry out the test away from light, and prepare the test solution and the reference solutions immediately before use.
Carry out the test through thin-layer chromatography (V.6.20.2), by using a plate coated with HF254 R silica gel.
Solution to be tested. Dissolve 50 mg vincamine SCR fr in chloroform R and dilute to 5 ml with the same solvent.
Reference solution (a). Dissolve 0.1g vincamine SCR fr in chloroform R and dilute to 10 ml with the same solvent.
Reference solution (b). Dissolve 20 mg vincamine SCR fr in chloroform R and dilute to 100 ml with the same solvent.
Reference solution (c). Take 5 ml reference solution (b) and dilute to 20 ml with chloroform R.
Put 10 l of each solution separately on the plaque. Develop over a path of 12 cm, with a mixture of 90 volumes of chloroform R and 10 volumes of methanol R. Let the plaque dry in air. Examine under ultraviolet light at 254 nm, then spray the reagent with iodoplatinate R.
Epivincamine. If there is a spot, which corresponds to the epivincamine in the chromatogram obtained with the test solution, it is not more intense than the principal spot in the chromatogram obtained with reference solution (b).
Other substances. If there are other spots besides the principal spot and the spot corresponding to the epivincamine in the chromatogram obtained with the solution to be examined, none of them is more intense than the principal spot in the chromatogram obtained with reference solution (c).
Loss on desiccation (V.6.22). Carry out on 1.00g vincamine in a heating cabinet at 100-105°C. The loss on drying is not more than 0.5%.
Sulphated ash (V.3.2.14). Carry out on 0.5g vincamine. The rate of sulphated ash is not more than 0.2%.
MEASUREMENT
Dissolve 0.300g vincamine in the mixture of 25ml anhydrous acetic acid R and 5ml acetic anhydride R. Carry out the measurement of the bases in a non-aqueous environment (V.3.5.5) by titrating through 0.1N perchloric acid in the presence of 0.2ml crystal violet solution R till the color turns to pure green. Carry out a titration till the color turns white.
1ml 0.1N perchloric acid corresponds to 35.44 mg C21H26N2O3 .

Store in a properly closed container, protected from light.
