Source text - English Common sense directs us to understand that there is a relationship between shaping and filling. If the canal space was larger, then it would most likely be more thoroughly debrided during the shaping procedure. If the canal shaping included a greater taper than 0.02 mm/mm, then the gutta-percha points placed into the canal and subject to both vertical and lateral condensation could also have a greater taper, providing greater resistance form and being far less likely to be driven over the apex. If the canals had a greater taper, then they would also be much easier to irrigate. Tissue-dissolving solutions would be more likely to come into closer approximation with the dentinal tubules, which would also be more open once the smear layer was removed with EDTA

The recommended dose of XXX is 1 mg/kg body weight administered once every 2 weeks as an intravenous infusion.
Lower dosing regimens have been used in clinical studies. In one of these studies, performed in adult male patients, after an initial dose of 1.0 mg/kg every 2 weeks for 6 months, 0.3 mg/kg every 2 weeks may maintain clearance of GL-3 in certain cell types in some patients; however, the long-term clinical relevance of these findings has not been established (see section 5.1).
The initial infusion rate should be no more than 0.25 mg/min (15 mg/hour) to minimize the potential occurrence of infusion-associated reactions. After patient tolerance is established, the infusion rate may be increased gradually with subsequent infusions.
Infusion of XXX at home may be considered for patients who are tolerating their infusions well. The decision to have a patient move to home infusion should be made after evaluation and recommendation by the treating physician. Patients experiencing adverse events during the home infusion need to immediately stop the infusion process and seek the attention of a healthcare professional. Subsequent infusions may need to occur in a clinical setting. Dose and infusion rate should remain constant while at home, and not be changed without supervision of a healthcare professional.

Рекомендована доза препарату ХХХ становить 1 мг/кг маси тіла, яка вводиться один раз на 2 тижні у вигляді внутрішньовенної інфузії.
У клінічних дослідженнях використовувалися нижчі дози. В одному з таких досліджень, проведеному у дорослих чоловіків, після початкової дози 1,0 мг/кг, яка вводилася кожні 2 тижні протягом 6 місяців, дозування 0,3 мг/кг кожні 2 тижні могло підтримувати зниження концентрації GL-3 в певних типах клітин деяких пацієнтів; проте клінічна значущість цих даних у довгостроковій перспективі не встановлена (дивіться розділ 5.1).
Для зведення до мінімуму потенційного ризику виникнення реакцій, пов’язаних з інфузією, початкова швидкість інфузії не повинна перевищувати 0,25 мг/хв (15 мг/год). Після підтвердження нормальної реакції пацієнта на препарат можна поступово збільшувати швидкість введення з кожною наступною інфузією.
Пацієнтам, які добре переносять інфузії препарату ХХХ, можна дозволити робити їх вдома. Рішення про переведення пацієнта на домашній режим проведення інфузій приймається після оцінки стану пацієнта лікарем та з урахуванням наданих ним рекомендацій. Пацієнти, у яких спостерігатимуться будь-які побічні явища при проведенні інфузій у домашніх умовах, повинні негайно припинити процес інфузії та звернутися за допомогою до медичного працівника. Можливо, що наступні інфузії необхідно буде здійснювати в медичному закладі. При введенні препарату в домашніх умовах його доза та швидкість інфузії повинні залишатися незмінними; пацієнту не можна змінювати їх самостійно, без нагляду медичного працівника
