The present invention relates to a fluororubber formed product having excellent mechanical properties at high temperatures.
BACKGROUND ART
Fluororubbers are known to be excellent in chemical resistance, oil resistance, and heat resistance, and also to have good compression set resistance at high temperatures. Fluororubbers are now desired to have better mechanical properties at high temperatures, such as strength at high temperature and elongation at high temperature. For example, when a cross-linked fluororubber product is used at as high temperature as more than 100°C, the product is required to have excellent mechanical properties at high temperatures as well as heat resistance, for high durability.
In terms of an increase in the compression set resistance, compositions such as one taught in Patent Document 1 have been proposed. Those compositions, however, have low elongation at room temperature, and therefore will probably have lower elongation at high temperature. The composition described in Patent Document 2 has higher elongation at high temperature, but does not have resistance to more severe use environment. The combination of a fluororubber and a thermoplastic fluoroelastomer in Patent Document 3 is an example of higher strength at high temperature, but the elongation at room temperature of this composition is also low, and therefore the elongation at high temperature will probably be even lower.
Document WO 2008/078738 A1 discloses compositions of fluororubber having more than 10 mol-% of TFE and MT Carbon Black, having a N2SA value of 8 m2/g. Similar carbon black is used in document EP 2 264 100 A1. Document JPH 03 122153 A discloses chemical, heat and oil resistant fluororubbers, having improved tensile strength, prepared using carbon black having a higher nitrogen adsorption specific surface area, but does not disclose fluororubbers comprising units derived from a compound of Formula (1) or a bis-olefin, and 0-10 mol-% of TFE units.

[0002] È noto che le fluorogomme hanno un’eccellente resistenza chimica, resistenza agli oli e resistenza termica, e inoltre hanno una buona resistenza alla deformazione da compressione interna a temperature elevate. Ora si desidera che le fluorogomme abbiano desiderato proprietà meccaniche migliori a temperature elevate, come resistenza a temperatura elevata e allungamento a temperatura elevata. Per esempio, quando un prodotto di fluorogomma reticolato viene usato temperatura elevate fino a più di 100 °C, per un’elevata durevolezza è necessario che il prodotto abbia eccellenti proprietà meccaniche a temperature elevate nonché resistenza termica.

La combinazione di una fluorogomma e un fluoroelastomero termoplastico nel documento brevettuale 3 rappresenta un esempio di una resistenza a temperatura elevata superiore, tuttavia l’allungamento a temperatura ambiente di questa composizione è anch’esso ridotto, e pertanto anche l’allungamento a temperatura elevata sarà probabilmente inferiore.
[0004] Il documento WO 2008/078738 A1 descrive composizioni di fluorogomma aventi più del 10% in moli di TFE e nerofumo MT avente un valore N2SA di 8 m2/g. Un nerofumo simile viene usato nel documento EP 2 264 100 A1. The present invention relates to a recombinant protein having the activity derived from the α-subunit of wild-type human β-hexosaminidase and the resistance to protease.
BACKGROUND ART
Both of Tay-Sachs disease and Sandhoff disease are diseases exhibiting neurological symptoms which are caused by accumulating GM2 ganglioside in neural system cells due to decreased activity of β-hexosaminidase A (Hex A). Hex A is a heterodimer composed of an α-subunit and a β-subunit and has the enzyme activity to degrade GM2 gangliosides. Tay-Sachs disease is a Hex A deficiency caused by α-subunit deficiency and Sandhoff disease is a Hex A deficiency caused by β-subunit deficiency.
The present inventors have previously provided a cell line which was established by introducing an expression vector into which genes encoding an α-subunit and a β-subunit (HEXA cDNA and HEXB cDNA, respectively) are inserted, into CHO cell strains or specific yeast strains and which constitutively expressed a wild-type recombinant Hex A. The present inventors found that the administration of thus produced wild-type recombinant Hex A to Sandhoff disease model mice leads to the reduction of GM2 gangliosides accumulated in neural system cells and the improvement of neurological symptoms, and thus confirmed that an enzyme replacement therapy is efficacious for Tay-Sachs disease and Sandhoff disease (Patent Document 1 and Non-Patent Document 1).
However, in general, in the case where a therapeutic agent containing an enzyme which is deficient is repeatedly administered to a patient, the enzyme in the therapeutic agent is often recognized as a foreign matter in the patient and thus the antibody is produced. Consequently, adverse/side reactions such as allergy reaction and anaphylactic reaction are occurred in the patient. Therefore, in the case where a wild-type recombinant Hex A is directly administered to a patient with Tay-Sachs disease or Sandhoff disease, adverse/side reactions may be occurred in the patient similarly as described above. Furthermore, a wild-type recombinant Hex A has disadvantages such as low stability in blood (plasma) and low ratio of uptake into cells of a disordered organ (neural system cells).
In order to solve these problems, the present inventors produced a modified β-subunit in which the active site of a β-subunit is substituted with the active site of an α-subunit based on conformational information of the α-subunit and β-subunit of β-hexosaminidase. Furthermore, the present inventors produced a modified β-hexosaminidase B which is a homodimer composed of the modified β-subunit (hereinafter, referred to "ModB") and confirmed that the recombinant enzyme has the activity to degrade GM2 gangliosides (Patent Document 2 and Non-Patent Document 2).

CONTESTO DELLA TECNICA
Sia di morbo di Tay-Sachs sia il morbo di Sandhoff sono malattie che presentano sintomi neurologici che sono causati dall’accumulo di ganglioside GM2 nelle cellule del sistema neurale a causa dell’attività diminuita della β-esosaminidasi A (Hex A). Hex A è un eterodimero composto da una subunità α e una subunità β e ha l’attività enzimatica di degradare i gangliosidi GM2. Il morbo di Tay-Sachs è una deficienza di Hex A causata da una deficienza di subunità α e il morbo di Sandhoff è una deficienza di Hex A causata da una deficienza di subunità β.
I presenti inventori hanno fornito in precedenza una linea cellulare, la quale è stata creata introducendo un vettore di espressione nel quale vengono inseriti geni codificanti una subunità α e una subunità β (rispettivamente cDNA di HEXA e cDNA di HEXB), in ceppi di cellule CHO o specifici ceppi di lievito e la quale esprimeva in modo costitutivo una Hex A wild type ricombinante. I presenti inventori hanno riscontrato che la somministrazione di Hex A ricombinante wild type prodotto in questo modo a topi di un modello di morbo di Sandhoff porta alla riduzione di gangliosidi GM2 accumulati nelle cellule del sistema neurale e al miglioramento dei sintomi neurologici e, pertanto, hanno confermato che una terapia sostitutiva enzimatica è efficace per il morbo di Tay-Sachs e il morbo di Sandhoff (documento brevettuale 1 e documento non brevettuale 1).
Tuttavia, in generale, nel caso in cui un agente terapeutico contenente un enzima che è deficiente sia somministrato in modo ripetitivo a un paziente, l’enzima nell’agente terapeutico sovente viene riconosciuto come materia estranea nel paziente e pertanto viene prodotto un anticorpo. Di conseguenza, nel paziente si verificano reazioni avverse/collaterali, come una reazione allergica e una reazione anafilattica. Pertanto, nel caso in cui sia somministrata una Hex A ricombinante wild type direttamente a un paziente con il morbo di Tay-Sachs o il morbo di Sandhoff, possono verificarsi reazioni avverse/collaterali nel paziente, analogamente a quanto descritto sopra. Inoltre, una Hex A ricombinante wild type presenta svantaggi, come bassa stabilità nel sangue (plasma) e basso rapporto di captazione nelle cellule di un organo malato (cellule del sistema neurale).
Al fine di risolvere questi problemi, i presenti inventori hanno prodotto un’subunità β modificata nella quale il sito attivo di una subunità β è sostituito con il sito attivo di una subunità α in base a informazioni conformazionali della subunità α e della subunità β di β-esosaminidasi. Inoltre, i presenti inventori hanno prodotto una β-esosaminidasi B modificata che è un omodimero composto dalla subunità β modificata (di seguito denominato “ModB”) e hanno confermato che l’enzima ricombinante ha l’attività di degradare i gangliosidi GM2 (documento brevettuale 2 e documento non brevettuale 2).
