After estimating long-run and short-run buoyancy, we explore, in this section, whether the stabilization role of taxation (captured by the short-run buoyancy) has varied during periods of economic expansion and economic contraction. For example, empirical evidence suggests that compliance falls quickly and quite strongly, although for a short time, as output falls below potential. This would imply that the response to revenue to changes in GDP would differ depending on whether output is falling below potential, or increasing above potential (IMF, 2015, and Sancak and others, 2010).
We begin with an event-study type exercise, by plotting the overall tax revenue to GDP ratios by country group around recession episodes (identified as years of negative GDP growth, year t in the graphs of Figure 4, below), as well as one and two years before/after the episode.

The first observation coming out of Figure 3 is that overall tax revenues decrease immediately from t-1 to t in all country groups, that is, the short-run buoyancy of tax revenues appears to be greater than one during a recession, suggesting that the tax system have been operating as an output stabilizer in the year of a recession. That said, in emerging and low income countries tax revenues seem to recover immediately in t+1, while it generally takes some time for that to materialize in advanced economies, where tax revenues remain subdued for a longer period. Looking at the four different tax categories, such lagged effect in advanced economies can be explained by a lagged and slow recovery in personal income taxes and taxes on goods and services. Social security contributions seem to increase in the year of the recession across all groups.
To further explore whether the tax system is buoyant during recessions, we estimate equation (3) only over 2008-2012, a period commonly identified as associated to the Global Financial Crisis. Results (displayed in Table A4 in the Appendix 1) suggest that, relative to estimates presented in Table 3, most of the buoyancy estimates (when statistically significant) are higher in magnitude, which indicates that after the Great Recession tax systems behaved slightly more as output stabilizers than on average in normal times. We also repeat the previous exercise, but now for each tax category for the reduced 2008-2012 period. Evidence (displayed in Table A5 in the Appendix 1) suggests that in advanced economies, PIT and TGS long run buoyancy coefficients are not only larger during the GFC relative to the average value, but they are also significantly larger than one. In the case of short-run buoyancy all taxes saw their contributions as output stabilizers enhanced during the Great Recession, in both AEs and EMEs. For low income countries such contribution came from CIT and TGS.
To formalize the above findings and in order to better explore whether tax buoyancy varies depending on the phase of the business cycle, the following alternative short-run regression is estimated:

where z is an indicator of the state of the economy (using the real GDP growth rate) normalized to have zero mean and unit variance. The remainder of the variables and coefficients are defined as before. This approach is equivalent to the smooth transition autoregressive (STAR) model developed by Granger and Teravistra (1993). The main advantage of this approach relative to estimating SVARs for each regime is that it considers a larger number of observations to compute the impulse response functions, thus making the responses more stable and precise.
Table 8 shows that in advanced countries only CIT buoyancy is larger during contractions than during times of economic expansions. Hence, this tax category on average seems to work better as output stabilizer during bad times compared to good times. In emerging market economies, this effect is attributed to both CIT but also to TGS, the buoyancy coefficients of which are found to be statistically larger than one during periods of relatively low growth. Overall, this result is confirmed by Furceri and Jalles (2016) who find that the overall impact of fiscal stabilization (measured as the amount of counter-cyclicality) is larger during recessionary periods.

Monoclonal antibodies have very complex structures, both chemically and physically, to maintain their functionality and stability during their lifetime.7 The native structures of mAbs and their domains have to adapt to changes in factors such as ionic strength, pH, and others. While mAb flexibility is necessary for biological function, it presents many challenges in formulation development. Under various conditions of a development process, the conformation, protein-solution interactions, and even chemical structure of a mAb can change, creating new variants with possible adverse effects on efficacy and stability. A full understanding of all possible degradation pathways under different stresses is still not complete, but many years of research by both academic and biopharmaceutical scientists have established the general scientific foundation for understanding protein properties in different environmental conditions.8,9 In formulation development, it is critical to have information on multidimensional properties of protein and their dependence on both formulation and stress factors. Different conditions can affect different aspects of protein stability.10 Generally, three types of stability can be distinguished. Conformational stability describes the ability of protein to maintain its native structure, to be properly folded. Colloidal stability is the stability of protein with native structure in solution to avoid aggregation, precipitation, or phase separations. Chemical stability is the stability of amino acids and the covalent bonds between them and between different protein domains.
Also conformational, colloidal, and chemical stabilities are not necessarily independent of each other. For example, protein unfolding can expose hydrophobic regions of a protein, leading to self-association often followed by total precipitation of the protein. Chemical modifications can create hydrophobic areas with the described above consequences. But a thorough knowledge of each stability contribution to overall stability properties can help significantly in the selection of the best formulation.
Conformational stability of monoclonal antibodies is high relative to other proteins. Normally, it is hard to achieve complete unfolding of an antibody molecule even at temperatures up to 100 °C. The technique of choice for measuring unfolding temperatures and thermodynamic parameters during protein unfolding is differential scanning calorimetry (DSC). A typical DSC profile for an IgG antibody molecule contains three peaks identified as Fab, CH2, and CH3 domains.11 The melting temperature (Tm) of each domain is not a constant but can be shifted depending on the formulation factors. Under typical formulation conditions, the most stable domain is usually the CH3 domain, and the least stable is the CH2 domain. The pH of the solution and the presence of salt significantly decrease the Tm.12 Some excipients can also change the Tm. The DSC method requires approximately 0.5 to 1.0 mL of sample at a protein concentration of 0.5 mg/mL for sufficient sensitivity, and the entire scan requires greater than 1 h to complete.
Instability under stress conditions can lead to structural changes, which otherwise require significant energy. The stress can be caused by elevated temperature, chemical agents, and surface or interphase interactions. On the other hand, monoclonal antibodies are very stable structurally under typical storage conditions at 4 to 8 °C for long periods of time. The major degradation pathway for these molecules under these conditions is aggregation or formation of large particles.23 Although aggregation of monoclonal antibodies after longterm storage at low temperature can sometimes be caused by unfolding,24 the major cause is colloidal instability. Generally, the structure of engineered mAb-based modalities is less stable than the original antibody molecule.
Screening for Colloidal Stability
Monoclonal antibodies are very complex proteins with multidomain structures. The most common IgG type contains 12 domains that can be divided into heavy and light chains, variable and constant domains. There are a total of four isotypes and two light chain classes expressed in humans. But those differences in isotype structures cannot explain the high variability in IgG stabilities.30 The mechanism of colloidal stability in biotherapeutic formulations is still poorly understood,31,32 despite an abundance of fundamental knowledge of physical processes in protein solutions. Only recently, systematic approaches have been applied to study the colloidal stability of mAbs in formulations. The approaches combine computational and experimental methods to understand protein-protein and protein-solution interactions, including mAb interactions with different formulation components. The most common computational method is calculation of the theoretical pI, which can serve as a reference value to estimate the stable pH range of potential formulations. At a pH close to the pI, the protein tends to precipitate from solution,33 and to avoid this, it is necessary to adjust the ionic strength by addition of salt or other excipients.34,35 For most mAb molecules, the pI range is between 6.0 and 9.0, but it is possible that various new mAb modalities may fall outside of this range.
In addition to formation of soluble aggregates and large particles, there is another manifestation of colloidal instability— phase separation. Massive precipitation and formation of insoluble large particles is one form of phase separation from the soluble protein fraction.
There are many methods for direct detection and measurement of protein aggregation. Antibody formulations stored at low temperatures for a long period of time can form soluble dimers, subvisible particles, and large visible particles. The process of native aggregation can be very fast and caused by significant and sudden changes in protein concentration, pH, or ionic strength. It can be slow, taking many months, and worst, it can also be unpredictable, induced by local fluctuations in surface properties. Formulation screening almost always includes sample analysis by SEC to measure the amount of soluble aggregates. The SEC method can detect mAb aggregates of sizes from dimers up to heptamers, with larger aggregates being filtered by the column. Modern SEC methods are based on ultra-performance liquid chromatography (UPLC) instruments, and the runtime for each sample can be as short as 2 to 6 min.
Screening for Chemical Stability
Chemical modifications of monoclonal antibodies can be predicted in many cases by analysis of the primary sequence.64 So-called hot spots of chemically unstable regions can be identified during early stages of development and, if possible, they can be engineered out. Higher order structural information is also critical and very helpful to determine the proximity of amino acid residues to each other and their exposure to the solvent, providing clues to the likelihood of chemical reactions occurring at given residues.65,66 Stress formulation conditions can create additional degradation pathways not readily predictable by computational methods. C-terminal processing of lysine residues,67,68 N-terminal pyroglutamate formation,69 deamidation,70 glycation,71 and oxidation72,73 are common chemical modifications of mAbs and their modalities.
Analysis of High-Throughput Formulation Data
The analysis of data starts with the planning of an experiment and the experimental design of that experiment. It is critical to collect all available information about the stability of a molecule before starting any formulation studies. Usually, initial results on protein purification and solubility are available even during early stages of development. Amino acid sequence, theoretical pI, and total charge can be used for “hot spot” analysis for potential chemical modifications and for estimates of physical stability. Furthermore, it is highly desirable to use basic quality by design (QbD) principles in the experimental planning.107,108 This means that it would be possible to predict the formulation properties from the results of a well-designed experiment. In formulation development, the combination of formulation components creates the formulation space, and the data obtained from stability studies create the formulation stability space. In cases where the number of formulations is too large to screen, a design of experiment (DOE) approach can significantly save limited resources. The data obtained from experiments designed by DOE methods can produce a model of output parameters (e.g., physical or chemical stability) that will cover formulation space not included in experimental conditions. With the tools of statistical analysis, one can estimate the significance of formulation factors and confidence intervals for the predictive models.
For example, 96 formulations of IgG1 and IgG2 were screened before and after incubation at different temperatures.99 Size exclusion chromatography was used to measure the percentage of monomer in each formulation, and a model was created with monomer percentage as the response value.

تحت شرایط فشار بی ثباتی می تواند منجر به تغییرات ساختاری شود که در غیر این صورت به انرژی قابل توجهی نیاز دارد. فشارمی تواند ناشی از درجه حرارت بالا، عوامل شیمیایی و فعل و انفعالات سطحی یا بین مرحله ایی باشد. از سوی دیگر، آنتی بادی های مونوکلونال در شرایط ذخیره سازی معمول در دمای 4 تا 8 درجه سانتیگراد برای مدت زمان طولانی به لحاظ ساختاری بسیار پایدار هستند. در این شرایط مسیر اصلی تخریب برای این مولکول ها تجمع یا تشکیل ذرات بزرگ است .23 اگرچه تجمع آنتی بادی های مونوکلونال پس از ذخیره طولانی مدت در دمای پایین می توانند گاهی اوقات با آشکار شدن ، بوجود آیند، 24اما علت اصلی آن بی ثباتی کلوئیدی است. به طور کلی، ساختار حالتهای طراحی شده مبتنی بر mAb از ثبات کمتری نسبت به مولکول اصلی آنتی بادی برخوردار است.
نمایش برای ثبات کلوئیدی
آنتی بادی های منوکلونال پروتئین بسیار پیچیده ای با ساختارهای چند دامنه ای هستند. شایع ترین نوع IgG حاوی 12 دامنه است که می تواند به زنجیره های سنگین و سبک، دامنه های متغیر و ثابت تقسیم شود. در مجموع چهار ایزوتایپ و دو طبقه زنجیره سبک وجود دارد که در انسان نشان داده شده است. اما تغییرات بالا در استحکامات IgG نمی توانند این تفاوت ها در ساختارهای ایزوتوپی را نشان دهند. 30 علی رغم دانش بنیادی بسیار از فرایندهای فیزیکی در محلول های پروتئینی ، 31 و 32هنوز مکانیسم ثبات کلوئیدی در فرمولاسیون های بیو تراپی بدست نیامده است. تنها اخیرا روشهای نظامندی برای مطالعه ثبات کلوئیدی از mAbs در فرمولاسیون ها استفاده شده است. رویکردهای مذکور روشهای محاسباتی و تجربی را ترکیب می کنند تا فعل و انفعالات پروتئین- پروتئین و پروتئین- محلول، از جمله فعل و انفعالات mAb با اجزای فرمولاسیون مختلف را بفهمند.
رایجترین روش محاسبه، محاسبه PI نظری است که می تواند به عنوان یک مرجع برای تخمین محدوده PH ثابت از فرمولاسیون ممکن استفاده شود. در pH نزدیک به PI، پروتئین معمولا در محلول رسوب می کند 33 و برای جلوگیری از این، لازم است که قدرت یونی را با افزودن نمک یا سایر امولسیون ها تنظیم کرد. 34،35 برای بیشتر مولکول های mAb، محدوده pI بین 6.0 و 9.0 است ، اما این امکان وجود دارد که حالتهای مختلف جدید mAb ممکن است خارج از این محدوده باشد.
علاوه بر شکل گیری رسوبات قابل حل و ذرات بزرگ، نمونه ی دیگری از جدایی فاز- بی ثباتی کلوئیدی وجود دارد. رسوب عظیم و تشکیل ذرات بزرگ غیر قابل حل نمایی از جداسازی فاز از کسر پروتئین محلول است.
روش های متعددی برای تشخیص مستقیم و اندازه گیری تجمع پروتئین وجود دارد. فرمولاسیون آنتی بادی های ذخیره شده در دماهای پایین برای مدت زمان طولانی می تواند دیمرهای محلول، ذرات نامرئی و ذرات قابل مشاهده را تشکیل دهد. فرایند تجمع اولیه می تواند بسیار سریع ، و ناشی از تغییرات قابل توجه و ناگهانی در غلظت پروتئین، pH یا قدرت یونی باشد. این می تواند کند، در حالیکه چند ماه طول می کشد ، و بدتر از آن، همچنین می تواند غیر قابل پیش بینی و ناشی از نوسانات اولیه در ویژگیهای سطحی باشد. نمایش فرمولاسیون تقریبا همیشه شامل تجزیه و تحلیل نمونه هایی با SEC برای اندازه گیری مقدار رسوبات محلول است. روش SEC میتواند مجموع اندازههای mAb از dimers(دیمر) تا heptamers ( هپتامر) را با مجموع های بزرگتر شناسایی کند که از طریق ستون فیلتر شده اند. روش های نوین SEC بر اساس دستگاه های کروماتوگرافی مایع در فشار بالا (UPLC) است و زمان مصرف برای هر نمونه می تواند تا 2 تا 6 دقیقه کوتاه باشد.
نمایش برای ثبات شیمیایی
حالتهای شیمیایی آنتی بادی های مونوکلونال می توانند در بسیاری موارد در تجزیه و تحلیل مراحل اولیه پیش بینی شوند. 64 نقاط به اصطلاح داغ مناطق ناپایدار شیمیایی می توانند درطی مراحل اولیه ایجاد شناسایی شده و در صورت امکان می توانند در خارج ساخته شوند. به منظور تعیین مجاورت باقی مانده های آمینو اسیدها با یکدیگر و قرار گرفتنشان با حلال اطلاعات ساختاری منظم نیز بسیار مهم و حیاتی هستند، در حالیکه نشان هایی را برای احتمال واکنش های شیمیایی موجود در باقی مانده های خاصی فراهم می کنند. 65،66 شرایط تشکیل فشار می تواند مسیرهای مخرب اضافی ایجاد کند که به راحتی قابل پیش بینی با روش های محاسباتی نیستند. تولید c-terminal از باقی مانده های لایسین (lysine)، 67و 68 تشکیلN-terminal متیلاسیون ، 69آلیسین ، 70 گلیکاسیون ، 71 ، 72و 73 اکسیدین ، تغییرات رایج شیمیایی از mAbs و حالتهای آنها هستند.
تجزیه و تحلیل داده های فرمولاسیون با کارایی بالا
تجزیه و تحلیل داده ها با برنامه ریزی یک آزمایش و طراحی تجربی آن آزمایش شروع می شود. قبل از شروع هر نوع مطالعات فرمولی، جمع آوري اطلاعات موجود در مورد ثبات مولکول مهم است. معمولا نتایج اولیه در تصفیه و حلالیت پروتئین حتی در طی مراحل اولیه موجود هستند. توالی آمینو اسیدها، pI نظریه، و طیف کامل می تواند برای تجزیه و تحلیل " نقطه جوش" استفاده می شود تا تغییرات شیمیایی و ثبات فیزیکی را بررسی کرد. بعلاوه، استفاده ازاصول ارزیابی کیفیت (QbD) در برنامه ریزی تجربی بسیار مطلوب است. 107و 108 بدین معنا که می توان ویژگی های فرمولاسیون از نتایج آزمایش به خوبی طراحی شده را پیش بینی کرد. درایجاد فرمولاسیون ، ترکیبی از اجزای آن فضای فرمول را ایجاد و داده های تهیه شده ازمطالعات ثبات، فضای ثبات فرمول سازی را بوجود می آورند. در مواردی که تعداد فرمولاسیون ها برای نمایش روی صفحه بسیار زیاد است، طرحی از روش آزمایش (DOE) می تواند به طور قابل توجهی منابع محدودی را حفظ کند. داده های بدست آمده از آزمایشات طراحی شده با روش DOE می تواند مدلی از پارامترهای خروجی مانند ثبات فیزیکی و شیمیایی را تولید کند که فضای فرمول را پوشش خواهد داد نه اینکه شرایط آزمایشگاهی را در بر گیرد. با ابزارهایی از تجزیه و تحلیل ، می توان اهمیت عوامل فرمولاسیون و فواصل اطمینان برای مدل های پیش بینی را مورد ارزیابی قرار داد. به عنوان مثال، 96 فرمول های lgG1 و lgG2 در دماهای مختلف قبل و بعد انکوباتور یا دوره ی کمون مورد بررسی قرار گرفت. 99 برای اندازه گیری درصد مونومر در هر فرمولاسيون اندازه گیری کروماتوگرافی استفاده شده و مدل با درصد مونومر به عنوان مقدار پاسخ ایجاد شده است.
