Source text - English She had barely stepped into the house when she heard a distinct male voice. “Bunny,” she called in her sternest tone.
“In here!” Bunny sang out.
Kate tossed her jacket onto the hall bench and went into the living room. Bunny was sitting on the couch, all frothy golden curls and oh-so-innocent face and off-the-shoulder blouse far too lightweight for the season; and the Mintz boy from next door was sitting next to her.
This was a new development. Edward Mintz was several years older than Bunny, an unhealthy-looking young man with patchy beige chin whiskers that reminded Kate of lichen. He had graduated from high school two Junes ago but failed to leave for college; his mother claimed he had “that Japanese disease.” “What disease is that?” Kate had asked, and Mrs. Mintz said, “The one where young people shut themselves in their bedrooms and refuse to go on with their lives.” Except that Edward seemed bound not to his bedroom but to the glassed-in porch that faced the Battistas’ dining-room window, where day in and day out he could be seen sitting on a chaise longue hugging his knees and smoking suspiciously tiny cigarettes.
Well, all right: no danger of romance, at least. (Bunny’s weakness was football types.) Still, a rule was a rule, so Kate said, “Bunny, you know you’re not supposed to entertain when you’re on your own.”
“Entertain!” Bunny cried, making her eyes very round and bewildered. She held up a spiral notebook that lay open on her lap. “I’m having my Spanish lesson!”
“You are?”
“I asked Papa, remember? Señora McGillicuddy said I needed a tutor? And I asked Papa and he said fine?” “Yes, but . . .” Kate began.
Yes, but he surely hadn’t meant some pothead neighbor boy. Kate didn’t say this, however.(Diplomacy.) Instead, she turned to Edward and asked, “Are you especially fluent in Spanish, Edward?”
“Yes, ma’am, I had five semesters,” he said. She didn’t know whether the “ma’am” was smart-aleck or serious. Either way, it was annoying; she wasn’t that old. He said, “Sometimes, I even think in Spanish.”
This made Bunny give a little giggle. Bunny giggled at everything. “He’s already taught me so much?” she said.
Another irksome habit of hers was turning declarative sentences into questions. Kate liked to needle her by pretending she thought they really were questions, so she said, “I wouldn’t know that, would I, because I haven’t been in the house with you.”
Edward said, “What?” and Bunny told him, “Just ignore her?”
“I got an A or A-minus in Spanish every semester,” Edward said, “except for senior year, and that one wasn’t my fault. I was undergoing some stress.”
“Well, still,” Kate said, “Bunny’s not allowed to have male visitors when no one else is home.”
“Oh! This is humiliating!” Bunny cried.
“Tough luck,” Kate told her. “Carry on; I’ll be nearby.” And she walked out.
Behind her, she heard Bunny murmur, “Un bitcho”.
“Una bitch-AH,” Edward corrected her in a didactic tone.
They fell into a little spasm of snickers.
Bunny was not nearly as sweet as other people thought she was.
Kate had never quite understood why Bunny existed, even. Their mother—a frail, muted, pink- and-gold blonde with Bunny’s same asterisk eyes—had spent the first fourteen years of Kate’s life checking in and out of various “rest facilities,” as they were called. Then all at once, Bunny was born. It was hard for Kate to imagine how her parents had considered this to be a good idea. Maybe they hadn’t considered; maybe it had been a case of mindless passion. But that was even harder to imagine. At any rate, the second pregnancy had brought to light some defect in Thea Battista’s heart, or perhaps had caused the defect, and she was dead before Bunny’s first birthday. For Kate, it was hardly a change from the absence she’d known all her life. And Bunny didn’t even remember their mother, although some of Bunny’s gestures were uncannily similar—the demure tuck of her chin, for instance, and her habit of nibbling prettily on the very tip of her index finger. It was almost as if she had been studying their mother from inside the womb. Their aunt Thelma, Thea’s sister, was always saying, “Oh, Bunny, I swear, it makes me cry to see you. If you aren’t the image of your poor mother!”
Kate, on the other hand, was not in the least like their mother. Kate was dark-skinned and big-boned and gawky. She would have looked absurd gnawing on a finger, and nobody had ever called her sweet.
Kate was una bitcha.

Да, но он точно не думал, что репетитором будет сосед-травокур. Этого Кейт, конечно, не сказала (из чувства такта). Вместо этого она повернулась к Эдварду и спросила:
– Эдвард, ты так хорошо знаешь испанский?
– Да, мэм, я его учил два с половиной года в школе.
Кейт не поняла, было это «мэм» сказано с издёвкой или всерьёз. Её это в любом случае раздражало – не такая она и старая.
Эдвард добавил:

English to Russian: Clostridium tetani (Tetanus) (an extract) General field: Medical Detailed field: Biology (-tech,-chem,micro-)
Source text - English If culture of Clostridium tetani is to be carried out it is best to use pre-reduced oxygen blood agar plates incubated under anaerobic conditions. These plates should then be taken to the bedside in an anaerobic jar and inoculated with any necrotic material from the wound and then placed back into anaerobic conditions thus minimizing the time that the organisms will be subjected to atmospheric oxygen. Two cooked meat dextrose broths should also be used and after inoculation if a red, hot nail is added to the broths then this will quickly reduce the oxygen level in the broths and aid the recovery of the organism. On blood agar medium Clostridium tetani grows as an extremely fine, swarming layer over the surface of the plate. The colonies are rarely more than 1mm in diameter, slightly raised and have a ground glass appearance with a filamentous edge. Non-motile variants can produce colonies lacking this filamentous edge. They will show incomplete or a-haemolysis initially, followed by complete haemolysis caused by the tetanolysin. In chopped meat dextrose broth the medium becomes turbid and shows gas production. If two cooked meat dextrose broths are inoculated, to aid in the identification, one can be heated to 80°C for 10 minutes. In this process the vegetative organisms are killed and only the spores will remain. If Clostridium tetani is present these spores can then be re-incubated to give a growth of the organism.
Clostridium tetani does not ferment lactose, maltose, fructose, arabinose, mannose or xylose, but does produce a greenish fluorescence in MacConkey's Media containing neutral red. Agglutination identification is possible and ten serotypes have been defined, although of little use clinically this may be useful in the investigation of an outbreak. In routine practice few attempts are made to culture Clostridium tetani; it is difficult to culture, a positive result does not indicate if the organism contains the toxin producing plasmid, and Clostridium tetani may be present without disease in patients with protective immunity. This paucity of information on the bacteria in vitro means very little is known about antimicrobial sensitivity patterns, important if resistance were to develop in Clostridium tetani. Similarly there have been very few attempts to quantify the toxin load and assess the prognostic significance of this. If large amounts are produced the toxin may be transported by blood and the lymphatics as well as by direct entry into nerve fibres, hence more rapid and wider dissemination of the effects of the toxin.
The toxin is encoded on a 75kb plasmid and transcribed as a single polypeptide with a molecular weight of 150000, the complete amino acid sequence of the toxin is known (19, 20, 21). The polypeptide undergoes post-translational cleavage into two subchains, the heavy and light chain linked by a disulphide bond. The carboxyl terminal part of the H chain mediates attachment to gangliosides (GD1b and GT1b) on peripheral nerves subsequently the toxin is internalized (12). It then is then moved from the peripheral nervous system to the central nervous system by retrograde axonal flow and trans-synaptic spread. The light chain of the toxin acts as a zinc metallopeptidase, which cleaves synaptobrevin (42), a single base pair mutation in the light chain abolishes this proteolytic activity (35). Synaptobrevin is an integral membrane component of synaptic vesicles, when cleaved these vesicles containing the inhibitory neurotransmitter g-aminobutyric acid (GABA) cannot fuse with the presynaptic membrane and release their contents into the synaptic cleft. The alpha motor neurons are therefore under no inhibitory control and undergo sustained excitatory discharge causing the characteristic motor spasms of tetanus. The toxin exerts is effects on the spinal cord, the brain stem, peripheral nerves, at neuromuscular junctions and directly on muscles. To what extent cortical and subcortical structures are involved remains unknown, certainly the toxin is a potent convulsant when injected into the cortex of experimental animals.
Translation - Russian Для культивирования C.tetani лучше всего использовать предварительно восстановленный кровяной агар. Чашки выдерживают в анаэробных условиях, затем достают из анаэростата, засевают свежим (только что взятым) раневым отделяемым и помещают обратно в анаэростат, благодаря чему бактерии минимально подвергаются воздействию атмосферного кислорода. Также используют две пробирки с мясным декстрозным бульоном: после посева в каждую пробирку помещают раскалённую докрасна иглу, что позволяет быстро удалить кислород из бульона и способствует росту микроорганизмов. На кровяном агаре C.tetani образует ветвящиеся колонии, которые покрывают агар тонким слоем и размер которых обычно не превышает 1 мм в диаметре. Колонии матовые, слегка возвышаются над поверхностью среды и имеют волокнистые края, которые могут отсутствовать у колоний неподвижных форм. У таких колоний гемолиз поначалу неполный (альфа-гемолиз), со временем под действием тетанолизина происходит полный гемолиз. Декстрозный бульон с рубленым мясом мутнеет, в нём образуется газ. При посеве материала в две пробирки с мясным декстрозным бульоном для более точной идентификации бактерий одну пробирку подогревают в течение 10 минут до 80°С. При этом погибают вегетативные формы, и выживают только споры. При обнаружении спор C.tetani их повторно инкубируют, чтобы вызвать рост бактерий.
C.tetani не разлагает лактозу, мальтозу, фруктозу, арабинозу, маннозу и ксилозу. В среде МакКонки с нейтральным красным наблюдается зеленоватое свечение. Возможна идентификация в реакции агглютинации. Различают 10 серотипов возбудителя столбняка. Их клиническое значение невелико, но может быть полезно при диагностике в случае вспышки заболевания. В обычной практике C.tetani, как правило, не культивируют: бактерии плохо растут на питательных средах, и положительный результат не позволяет с уверенностью сказать, что данные бактерии содержат плазмид, отвечающий за производство токсина. Кроме того, C.tetani могут присутствовать в организме человека и не вызывать болезнь, если иммунитет не ослаблен. Нехватка данных о бактериях in vitro означает, что нам очень мало известно об их чувствительности к антибиотикам. Эти знания были бы очень важны в том случае, если бы C.tetani развили невосприимчивость к антимикробным препаратам. Немногочисленны и исследования, направленные на определение величины токсической нагрузки и оценку прогностического значения этих данных. В больших дозах токсин может переноситься по кровеносным и лимфатическим сосудам, а также напрямую проникать в нервные ткани, что приводит к более быстрому и обширному проявлению признаков столбняка.
