Source text - English OBJECTIVE—To establish the role of the transcription factor Pax4 in pancreatic islet expansion and survival in response to physiological stress and its impact on glucose metabolism, we generated transgenic mice conditionally and selectively overexpressing Pax4 or a diabetes-linked mutant variant (Pax4R129W) in b-cells.
RESEARCH DESIGN AND METHODS—Glucose homeostasis and b-cell death and proliferation were assessed in Pax4- or Pax4R129W-overexpressing transgenic animals challenged with or without streptozotocin. Isolated transgenic islets were also exposed to cytokines, and apoptosis was evaluated by DNA fragmentation or cytochrome C release. The expression profiles of proliferation and apoptotic genes and b-cell markers were studied by immunohistochemistry and quantitative RT-PCR.
RESULTS—Pax4 but not Pax4R129W protected animals against streptozotocin-induced hyperglycemia and isolated islets from cytokine-mediated b-cell apoptosis. Cytochrome C release was abrogated in Pax4 islets treated with cytokines. Interleukin-1b transcript levels were suppressed in Pax4 islets, whereas they were increased along with NOS2 in Pax4R129W islets. Bcl-2, Cdk4, and c-myc expression levels were increased in Pax4 islets while MafA, insulin, and GLUT2 transcript levels were suppressed in both animal models. Long-term Pax4 expression promoted proliferation of a Pdx1-positive cell subpopulation while impeding insulin secretion. Suppression of Pax4 rescued this defect with a concomitant increase in pancreatic insulin content.
CONCLUSIONS—Pax4 protects adult islets from stress-induced apoptosis by suppressing selective nuclear factor-kB target genes while increasing Bcl-2 levels. Furthermore, it promotes dedifferentiation and proliferation of b-cells through MafA repression, with a concomitant increase in Cdk4 and c-myc expression.

Translation - Spanish OBJETIVO—Para establecer el papel del factor de transcripción Pax4 en la expansión y supervivencia del islote pancreático en respuesta al estrés fisiológico y su impacto en el metabolismo de la glucosa, generamos ratones transgénicos que, condicional y selectivamente, sobreexpresan Pax4 o una variante mutante ligada a diabetes (Pax4R129W) en las células beta.
DISEÑO EXPERIMENTAL Y MÉTODOS—Se evaluaron la homeostasis de la glucemia y la muerte y proliferación de las células beta en animales transgénicos con sobreexpresión de Pax4 o Pax4R129W, expuestos o no a estreptozotocina. También fueron expuestos a citoquinas islotes transgénicos aislados, y se evaluó la apoptosis mediante la fragmentación del ADN o por liberación del citocromo C. Se estudiaron los perfiles de expresión de genes implicados en proliferación y apoptosis, y marcadores de células beta, mediante inmunohistoquímica y RT-PCR cuantitativa.
RESULTADOS—Pax4 (pero no Pax4R129W) protegió a los animales de hiperglucemia inducida por estreptozotocina, y a los islotes aislados de apoptosis de las células beta mediada por citoquinas. La liberación de citocromo C quedó suprimida en islotes Pax4 tratados con citoquinas. En los islotes Pax4 los niveles de transcritos de interleucina-1b quedaron suprimidos, mientras que en los islotes Pax4R129W aumentaron junto con los niveles de NOS2. Los niveles de expresión de Bcl-2, Cdk4 y c-myc aumentaron en islotes Pax4, mientras que los niveles de transcritos de MafA, insulina y GLUT2 quedaron suprimidos en ambos modelos animales. La expresión a largo plazo de Pax4 promovió la proliferación de una subpoblación celular positiva para Pdx1 y provocó una disminución en la secreción de insulina. La supresión de Pax4 invirtió este defecto, con un incremento concurrente del contenido de insulina pancreática.
CONCLUSIONES—Pax4 protege los islotes adultos de la apoptosis inducida por estrés, suprimiendo selectivamente genes diana del factor nuclear-kB al mismo tiempo que incrementa los niveles de Bcl-2. Además, promueve la desdiferenciación y proliferación de células beta a través de la represión de MafA, con incremento simultáneo de la expresión de Cdk4 y c-myc.

Source text - English Shinji's memories of his arrival in Tokyo-3 were fixed in the first seconds of his arrival when a spider landed on his nose. It had been floating through the air, held aloft by a gentle breeze and by a tangle of webs that bore some resemblance to a parachute made by MC Escher. As he stepped off the train, the breeze blew it onto his face. Many people would have screamed or hurriedly slapped it off his face. Shinji simply blinked and stared at it.
The train doors closed and it pulled out of the station. Shinji ignored it, focused on the spider. It sat on his cheek and stared at him. He stared back in a contest rigged against him, since his opponent had no eyelids. Slowly, Shinji put down his suitcase and raised his hand, brushing the spider onto his other hand, then putting it down on the ground. It scampered away, and he smiled a little smile for the first time in days.
"Kind to animals?" a woman's voice asked.
Shinji blinked and looked up. A woman was staring at him through mirrored sunglasses. She didn't look like someone who would work for his father; she looked more like an idol singer, dressed in casual, but sexy clothing, with a figure of the kind that Shinji was just now starting to notice. It wasn't her clothing or her body that drew his attention, but rather the strange necklace she wore. The chain was made of gold, while the pendant mixed silver and bronze, tracing out a sort of broken five pointed star with a tongue of flame in the middle made of copper. Shinji normally wasn't much for abstract art, or any kind of art for that matter, but it caught his attention and wouldn't let it go.
Translation - Spanish Los recuerdos de Shinji de su llegada a Tokyo-3 se fijaron en los primeros segundos, cuando una araña aterrizó en su nariz. Había estado flotando por el aire, sostenida por una brisa suave y por un enredo de telaraña que tenía cierto parecido a un paracaídas diseñado por MC Escher. La brisa se la plantó en la cara nada más bajar del tren. Mucha otra gente hubiera gritado o se la hubiera quitado de la cara a manotazos. Shinji simplemente parpadeó y se la quedó mirando.
El tren cerró las puertas y salió de la estación. Shinji lo ignoró, concentrado en la araña. Ésta estaba plantada en su mejilla y le contemplaba fijamente. Él le devolvía la mirada en una competición amañada en su contra, ya que su oponente no tenía pestañas. Lentamente, Shinji dejó su maleta en el suelo y levantó la mano, empujando la araña hasta su otra mano y depositándola seguidamente en el suelo. Ella huyó correteando, y él sonrió un poco por primera vez en días.
“¿Te gustan los animales?” preguntó una voz de mujer.
Shinji parpadeó y levantó la mirada. Una mujer le contemplaba a través de unas gafas de sol de espejo.
