Se comunica a la Compañía arriba referenciada que el expediente de Registro antes mencionado, ha sido evaluado en el Servicio de Autorización de Antimicrobianos de la División de Productos Biológicos y Biotecnología de la Agencia Española de Medicamentos y Productos Sanitarios. Que no puede emitirse en este momento un informe satisfactorio al Comité de Evaluación de Medicamentos en los aspectos contemplados por esta División en el Modulo 3, antes de que los puntos que se recogen en esta comunicación sean resueltos.
Por ello, se informa a la Compañía en el plazo máximo de 1 mes contado desde el envío de este escrito, se deberá aportar la información solicitada (en formato electrónico) presentándola mediante Registro General o Telemático.
Una vez entregada la documentación, se ruega lo comuniquen (telefónicamente/e-mail) a la persona de contacto o a la Secretaria de esta División de Productos Biológicos y Biotecnología.
En el caso de que en el encabezado de este oficio se considere que se precisa una entrevista, se ruega a la Compañía que se comunique telefónicamente con la persona de contacto para los detalles de la misma. En el caso de que no figure la necesidad de una entrevista, se ruega, salvo situaciones muy excepcionales, que no se realicen comunicaciones con la persona de contacto.
Si en el plazo anteriormente mencionado no se recibe respuesta de Ia Compañía contestando a los puntos solicitados, se informará por el procedimiento ordinario.
Se ruega encarecidamente a la Compañía receptora de esta comunicaci6n que siga del modo más riguroso las instrucciones antes aludidas y que cualquier duda sobre instrucciones generales del procedimiento sea consultada al Jefe de la División de Productos Biológicos y Biotecnología.

Para esta solicitud, se emitió un primer informe "Solicitud de aclaraciones y/o información adicional" fecha 2410612013 y una propuesta de denegación con fecha 911212013 señalando oportunamente las deficiencias encontradas.
La Compañía presenta alegaciones, respondiendo a las cuestiones mencionadas.
Estudiada de forma conjunta la documentación de soporte inicial y la adicional presentada, se solicitan las aclaraciones e información adicional siguientes:

La vigencia de la tabla compilada presentada en esta sección está supeditada a la adecuada actualización de las partes abiertas respectivas de los ASMFs y la posterior consideración y compilación de estos requerimientos e incluyendo además aquellos requisitos adicionales establecidos por el propio fabricante del PT como necesarios para garantizar la fabricaci6n y calidad del PT.
6. La tabla de especificaciones de API procedente y emitida por el fabricante de PT debe ser compilada y única y aplicada a la materia prima de forma indistinta de su origen.
Las especificaciones de materia prima establecidas por el fabricante de producto terminado deben incluir aquellos requisitos adicionales necesarios por motivos tanto de la fabricación como para garantizar el adecuado comportamiento del medicamento (liberación del API desde
la formulación).
Además, se han encontrado las siguientes deficiencias:
• El límite para metales pesados debe estar en consonancia con el límite establecido en la monografía de Ofloxacino de Farmacopea Europea.
• La frecuencia de control de algunos parámetros de la materia prima cada 5 lotes no se considera adecuada ni suficiente en este momento.
• Los límites de las Sustancias relacionadas deben ser únicos y aplicables
Independientemente de la fuente de API.

Es necesario garantizar inicialmente la calidad microbiológica en todos los lotes de manera rutinaria y solo cuando la experiencia en la producción haya sido demostrada debidamente, podrá solicitarse una menor frecuencia (mediante variación).

Ensayo microbiológico
La idoneidad del ensayo microbiológico ha sido proporcionada para Labor L +S AG Mangelsfeld 4 D-97708 Bad Bocklet-GroBenbranch.
12. Sírvanse confirmar cuales son las entidades responsables del control microbiológico rutinario conforme a la verificación aportada. Además deberá remitirse una declaración formal y el informe de la verificación llevada a cabo (procedente de la entidad(es) designada(s) al efecto).
Assay Method (HPLC) (incluyendo aspectos cuantitativos del ensayo de disolución)
13. Tras la revisión de las respuestas aportadas, debe quedar claro que el control de contenido (Assay) se realiza mediante HPLC-UV, incluyendo la determinación correspondiente de la fracción disuelta en el ensayo de disolución.
En el caso de que la determinación de contenido en el ensayo de disolución se realice por espectrofotometría UV (sin HPLC) el procedimiento debe quedar adecuadamente descrito y validado, separado de la información actualmente recogida para el HPLC-UV de Assay.
14. Validación (HPLC-UV). En el informe de validación se aprecian las siguientes deficiencias:
• Selectividad: el estudio con impurezas conocidas se sustenta sobre la base de cromatogramas en los que no se aprecia pico ninguno.
• Linealidad/rango: En consonancia las provisiones de Ia directriz Validation of analytical Procedures: text and methodology ICH Q2 (R1), se debería ajustar el rango de linealidad al 80-120 % de la concentración test.
3.2. P.8 Estabilidad
Protocolo:
15. Se deberá presentar el protocolo de estabilidad corregido de acuerdo a lo establecido en el presente informe. Los test que se van a llevar a cabo y sus límites, la frecuencia de cada análisis de acuerdo a las modificaciones que han surgido a lo largo de la evaluación de este trámite.
Resultados de estabilidad
Los estudios de estabilidad presentados como soporte del dossier corresponden a lotes fabricados en 2006-2009. Sin embargo en los resultados analíticos se presentan lotes de 2012. En consonancia con las NCF (GMP) deberían incluir en estabilidad los lotes de validación y a partir de ese momento
al menos 1/año.
16. Sírvanse aportar estudios de estabilidad sobre lotes fabricados después de 2009.

The company mentioned referenced above is informed that the Registration record mentioned before has been evaluated in the Authorization of Antimicrobial Service of the Biological Products and Biotechnology Division of the Spanish Agency for Medications and Health Products. That at this moment a satisfactory report cannot be issued to the Medications Evaluation Committee in the aspects covered by this Division in Module 3, before the points that are collected in this communication have been resolved.
For that reason, the Company is informed that within the maximum term of 1 month counting as of when this paper has been sent, it should supply the requested information (in electronic format) presenting it by means of a General or Telematic Record.
Once the documentation has been presented, we request that you inform (by telephone/e-mail) the contact person or the Secretariat of this Biological Products and Biotechnology Division.

The validity of the compiled table presented in this section is subject to the adequate updating of the respective open parts of the ASMFs and the later consideration and compilation of these requirements and also including those additional requirements established by the manufacturer itself of the FP as necessary for guaranteeing the manufacture and quality of the FP.
6. The API specification table from and issued by the manufacturer of the FP should be compiled and unique and applied to the raw material without distinction of it origin.
The specifications of the raw material established by the manufacturer of the finished product should include those additional requirements necessary for reasons of the manufacturing as well as to guarantee the adequate behavior of the medication (freeing the API from
the formulation).
Also, the following deficiencies have been found:
• The limit of heavy metals should be in line with the established limit in the Ofloxacin monograph of European Pharmacopeia.
• The control frequency of some parameters of the raw material each 5 lots is not considered adequate nor sufficient at this time.
• The limits of related Substances should be unique and applicable
Independently of the API source.

13. After the review of the answers provided, it should be clear that the quantity control (Assay) is carried out by means of HPLC-UV, including the corresponding determination of the dissolved fraction in the test dissolution.
In case the determination of the content in the dissolution test is carried out by UV spectrophotometry (without HPLC) the procedure should be adequately described and validated, apart from the information presently recollected for the Assay HPLC-UV.
14. Validation (HPLC-UV). In the validation report the following deficiencies were noticed:
• Selectivity: the study with known impurities is supported on the basis of chromatography in which no peak is noted.
• Linearity/rank: In line with the provisions of the Validation of analytical Procedures directive: text and methodology ICH Q2 (R1), the linearity rank should be adjusted to 80-120 % of the test concentration.

15. The stability protocol, corrected according to what is established in the present report, should be presented. The test that are going to be carried out and their limits, the frequency of each analysis according to the modifications that have emerged throughout the evaluation of this procedure.
Stability results
The stability studies presented as support for the dossier correspond to lots manufactured in 2006-2009. Nevertheless, in the analytical results lots from 2012 are presented. In line with the NCF (GMP) in stability the validation lots should be included and as of that moment
at least 1/year.
