Hello
I'm wondering if a glossary created in one language pair (ES-FR, for instance) could be used in a project with the opposite direction (FR-ES in this case). Or would I be forced to edit the columns manually by exporting the glossary to an Excel file, change their location and then to a txt file again?

Glossaries work only in one direction

Yes, text glossaries work in one direction only, since OmegaT has to rely on the column position to identify the source and target languages.
So, yes, you have to go through a spreadsheet program to invert the column location. You might find it more convenient to maintain the glossary in the spreadsheet program, and only use the text versions as export files.
The only glossary format working in both directions is TBX.
