Se toarnă în fiecare cutie Petri câte cca. 15 cm2 mediu PCA (Plate Count Agar) cu temperatura de 45±5°C. Mediile se uniformizează rapid în plăci, prin rotirea plăcilor în plan orizontal, apoi sunt lăsate în repaus până se produce solidificarea lor. Dupa solidificarea mediului plăcile se termostatează cu capacul în jos, pentru 3-5 zile la temperatura de 25-28°C.
