This reading just occurred to me - because earlier in the Will, the testator refers to his son as “er”( not “es”)


Ich versuch es mal auf Hochdeutsch: Hiermit vermache ich meinem Sohn Johannes meinen Hof, auf dem dieser zur Zeit lebt. Ich verfüge, dass mein Sohn Johannes für den oben benannten Hof 600 Pfund entrichten soll…. wobei diese Summe zu gleichen Teilen unter meinen fünf Töchtern zu verteilen ist. […] Und hiermit bestätige ich, dass meinen drei oben namentlich genannten Söhnen das oben bezeichnete Land zu ihrem vollständigen Eigentum übertragen werden soll, und zwar mit den gleichen Verfügungsrechten wie ich sie habe.
Ferner verfüge ich, dass mein Sohn Johannes die für diese Immobilie übliche Miete in Höhe von 100 Talern zahlen muss. (to whom exactly is not explained).
If read this way, Johannes gets the farm but has first to pay off his sisters and then also pay 100 Thaler (monthly???) because this is the current market rent.

I would agree with MalteLaurids, Cilian and your own suggestion. It's the 'es' part I don't get, because it's hard to say who it refers to. Btw, If the daughters get the 600 Pfundt, who actually gets the 100 Thaler? The sons?

I also thought of this as rent to be paid by Johannes retroactively. In this case "Rend" is rent and "was es auf dem Platz Gewohnet hat" would be something like "since he has lived on this land".
Another possibility could be that Johannes is supposed to pay some kind of compensation (as you said "Rend"=Rente) to someone who has lived there so far: "pay to those who have lived on the land".
However, I would imagine that there would be some mention of such person(s), like a name or relation or similar, so I tend to favor the first solution.

Since the testator is leaving his son Johannes the farm, on which he (Johannes) is now living, perhaps the questionable sentence is something like: "It is my will that my son Johannes is to pay the sum of one hundred dollars in rent for the time he has lived on the place..." Just a guess here!

I'm not really sure how "rent" is involved here. Earlier in the will, the testator writes: "Es ist mein Wille, uebergebe und vermache auf Immer an meinen sohn Johannes meine Plantasche...wo er jetz darauf wohnet. Es ist mein Wille das mein sohn Johannes Geben Soll vor obige Plantasche die Summa von sechs hundert Pfundt...es soll Gleich unter meine fuenf Toechter in Gleiche theil Getheilt werden." Later in the will, the testator states: "...und es ist mein Wille und bestaetige hiermit das meinen trey obengemelde soehne vor obengemeltes land Ihr Recht so Gut und vollstaendig sein soll als ich es habe." So he's basically SELLING his son Johannes full rights to the land, making him pay 600 pounds for it, which is to be divided among the five daughters. If Johannes gets full rights to the land, I'm stumped as to the comment concerning "Rend" (=Rente?).
