I often get mails like this one below:

Can you have it finished by Monday end of day?
Can you have it finished by Monday?

Could you guys please explain to me the meaning of preposition "by".

If someone tells you they want something "by Monday," it is acceptable to send it on Monday. You don't have to send it on Sunday (the day before). "By Monday" means that Monday is the deadline (the latest date you should send the work). By Monday "at the end of the day" implies the end of the workday, which could vary according to the country. It could also be later during the evening if an understanding has been reached. I would not ever assume that it is okay to send something at night, however, unless you have specifically discussed that with the client.
