Explanation:

Validacijsko ispitivanje bioanalitičkih postupaka uključuje odreñivanje u obogaćenim uzorcima («spiked» samples) pripravljenim iz referentnih standarda i biološkog matriksa te odreñivanjem relevantnih parametara koji opisuju izvedbu metode (1). Valja procijeniti ispravnost, preciznost, funkciju odgovora i selektivnost metode na endogene supstancije, metabolite i razgradne produkte u biološkom matriksu (27).

Obogaćeni/standardni/QC uzorci (1, 27). - Unutar validacijskog postupka koristi se isti biološki matriks za pripravu obogaćenih uzoraka, kalibracijskih standarda i uzoraka kontrole kvalitete («quality control», QC), koji se svi pripravljaju iz odvojenih odvaga. Svi se uzorci obrañuju na isti način. Biološki matriks treba takoñer validirati te navesti korišteni antikoagulans ili anestetik za dobivanje kontrolnog matriksa, materijal posuña, etc.

Obogaćeni uzorci («spiked samples», to su slijepi matriksi ili stvarni uzorci obogaćeni analitom) pripravljaju se iz kalibracijskih standarda (uzorci poznate čistoće, stabilnosti i uvjeta čuvanja: pripravljeni svježi svaki dan u istom matriksu).

Matrix Spike (Spiked Sample or Fortified Sample)– A sample prepared by adding a known mass of target analyte to a specified amount of matrix sample for which an independent estimate of target analyte concentration is available. Matrix spikes are used, for example, to determine the effect of the matrix on a method's recovery efficiency. (ORD) (OW/EAD) (OPP) (OAR/OAQPS) (ORCR)

Matrix Spike (MS) and Matrix Spike Duplicate (MSD) – Two aliquots of the same environmental sample to which known quantities of the target analytes are added in the laboratory. The MS and MSD are analyzed exactly like a sample, and their purpose is: to determine whether the sample matrix contributes bias to the analytical results, and to indicate precision associated with laboratory procedures. The background concentrations of the analytes in the sample matrix must be determined in a separate aliquot and the measured values in the MS and MSD corrected for background concentrations. (ORD)
