W kontekście inicjatywy przyjmuje się, że poważne transgraniczne zagrożenia zdrowia to zdarzenia wywołane chorobami zakaźnymi lub czynnikami biologicznymi powodującymi choroby niezakaźne, pochodzenia chemicznego, środowiskowego lub nieznanego bądź spowodowane zmianą klimatu, o potencjalnych poważnych skutkach dla zdrowia publicznego. Na potrzeby inicjatywy zagrożenia wynikające ze skutków zmiany klimatu będą rozpatrywane w zakresie zagrożeń środowiskowych. Zdarzenia radiologiczne nie wchodzą w zakres inicjatywy, ponieważ są objęte przepisami traktatu EURATOM.
