IC clearance may be affected by IC composition, particu-
larly when the antigen is also an antibody (thereby increasing
lattice size and speed of clearance). In examples 1 and 2 (pre-
sented subsequently), a mAb drug was administered to humans
(example 1) or monkeys (example 2), followed by a clearing
antibody (simulating an ADA-like response). In example 3, the
ICs were formed by the mAb drug complexing to its antigen in
vivo (IgE).
In example 1, an

I-labeled mouse anticancer mAb drug
was administered to patients, and in vivo IC formation and
clearance kinetics were measured (Davies et al. 1990). Patients
received 10 mg of the drug intraperitoneally (IP) on day 0,
followed by 18 mg of a clearance-accelerating antibody,

I-labeled human anti-mouse IgG, on days 1 and 2. The clear-
ing antibody was administered to promote IC formation/
clearance of the
131
I-labeled mouse anticancer mAb drug, thus
protecting the bone marrow from radiation. Following admin-
istration of the clearing antibody, large lattice (19–40S sedi-
mentation coefficient) ICs formed within 5 min, and were
cleared from plasma to the liver rapidly (half-life of 11 min).

Активные компоненты комплемента С4Ь и СЗЬ ковалентно связывают растворимые иммунные комплексы и через CR1 привязывают их к эритроцитам, которые уносят их с собой к макрофагам селезенки и печени, обеспечивая тем самым клиренс крови от иммунных комплексов. При этом
макрофаг «снимает» иммуный комплекс с эритроцита, не повреждая сам эритроцит. Если этот механизм клиренса крови от иммунных комплексов оказывается недостаточным, то «неубранные» комплексы выпадают в осадок.
