I couldn't find any references to fluctuations of medication effectiveness but quite a lot about motor fluctuations. Medication often loses its effectiveness with progression of the disease which causes motor fluctuations. At this point, motor fluctuations (also called “on-off” fluctuations) may begin to appear. They are designed to take effect quickly: within 15 minutes or less. Typically, the response is short, maybe an hour or two. It is called “rescue” therapy because it bridges gaps in other medication effectiveness.
