The primary purpose of this stage is to ensure that the laboratory’s analysts that will undertake the verification of the new method become fully familiar with all aspects of the method. This process is detailed in this section. The initial step would involve getting analysts used to performing the method using pure cultures of typical target bacteria defined by the scope of the test. Subsequently a broader range of strains of target and non-target bacteria are included to allow familiarisation with the range of typical positive and negative reactions that can be associated with the method (e.g., range of colony colours and morphology for membrane filtration methods or intensity of colour/fluorescence in tubes or wells for MPN methods). This is then followed by using natural samples known to contain the target bacteria typical of those normally analysed by the laboratory. This allows familiarisation with any interference from other bacteria or the sample matrix that may impact on the practicability of adopting the new method.
