RET is an abbreviation for "rearranged during transfection", as the DNA sequence of this gene was originally found to be rearranged within a 3T3 fibroblast cell line following its transfection with DNA taken from human lymphoma cells. The human gene RET is localized to chromosome 10 (10q11.2) and contains 21 exons.

Cabozantinib, which was approved by the FDA for the treatment of MTC in November 2012, inhibits the receptor tyrosine kinase for the RET (rearranged during transfection) proto-oncogene, as well as the kinases for MET and VEGFR2.

Approximately 75% of all cases of MTC occur sporadically, and of those cases, as many as 65% of patients have somatic RET mutations.
