"Them" refers back to "the different areas of design" in the first paragraph said by A.
He is probably taking about various challenges that he had to tackle in his works. Those may be specific technical issues, sometimes done by separate specialist designers. What he is saying is that he actually did those by himself, he feels lucky that he had the opportunity but also proud that he could do it.
He did not face those challenges just so he could say he has done them (ticking them off the list) but because he was really interested in them and intrigued by them, and wanted to develop himself, to reach higher in his profession.

At least this is what I get out of this conversation.

"Ticking them off the list" itself means accomplishing certain goals one by one - imagine you have a shopping list and as you go around the store you put items in your basket, and when you do that, you place a checkmark (tick) next to the items name. Same thing with a "To Do" list.
