The biological activities of CLA have received considerable attention because of its anti-cancer, anti-atherogenic and anti-diabetic effects. CLA prevents the development of atherosclerosis induced by a high cholesterol diet in rabbits. One possible mechanism is through inhibition of cyclooxygenase (COX-1 and COX-2), both of which have been implicated in the development of atherosclerosis. Recently, we have shown that selective inhibition of COX-1 inhibits the development but does not induce regression of atherosclerosis via a platelet dependent mechanism, whereas selective COX-2 inhibition has no effect. c9,t11- and t10,c12-CLA isomers are not substrates for COX but compete with arachidonic acid for access to the enzymes, therefore, preventing its metabolism to prostaglandins (PGs). Consequently, these CLA isomers inhibit platelet aggregation and PGE2 generation in macrophages, although the latter is in part due to suppression of COX-2.

Cyclooxygenase (COX) is an enzyme that is responsible for formation of important biological mediators called prostanoids (including prostaglandins, prostacyclin and thromboxane). Pharmacological inhibition of COX can provide relief from the symptoms of inflammation and pain; this is the method of action of well-known drugs such as aspirin and ibuprofen.
