Preparation of Hemophilus influenza type b vaccine.
Stage 2
Fermentation of Hib is done in fermentor with 30L capacity and working volume 20-24L. Fermentor is sterilized at 121 C for 30 minutes with Basal medium. Supplements are added by filter sterilization. The seed is used to inoculate the fermentor under aseptic conditions. Cultivation is performed at temperature 34-36 with agitation of 300-600 RPM, DO set point 20%, pH 7,1 for 8-12 hours with Glucose spike 1-2 of 20% each. OD590 is monitored on spectrophotometer. When it is >3,5 and stationary phase has reached, purity of broth is checked.

Under this operating strategy, the operator enters a low DO set point and a high DO set point through
the HMI. The blower runs until the high DO set point is reached. Once this has occurred, the blower
shuts down. When the low DO set point level is reached, the blower starts again.
