JESSE
Joey, I've got to talk to you.

JOEY
Babe, not now, I'm working the room. I'm the hostess with the mostess.

JESSE
No, you're a Hostess Twinkie.

I know what Twinkies are and I get "the host with the most" but what's really so funny about Jesse's reply?

Thanks!

Selected answer:You're an idiot.
Explanation:
Joey is brimming with confidence in anticipation of "working the room" and (he hopes) being popular with the ladies.

Jesse deflates his confidence by calling him a twinkie (play on words with "hostess" - Hostess is the company that created Twinkies), a fluffy brainless idiot....

Actually, Joey is acting like a real woman there (before this piece of text). So he's just satisfied with playing an important part in the party (others were doing other stuff like getting presents, etc).

But that doesn't really matter. Thanks for the insight!
