I'm looking for a suitable translation of "expediente" in the above quotation of an official letter from a government dept.

This is how it would be put in Business English in the US. "Numero deste expediente" reference to the reference number of the letter.

"Of this letter" doesn't really need to be included in the translation: Reference to an "above reference number" will be clear in and of itself.
