Source text - English Following sample preparation, recovered cells are automatically pipetted into multiple flowcell channels of the test cassette. Conductive layers of transparent indium tin oxide (ITO) coat the top and bottom inner surfaces of each flowcell channel and act as electrodes. An additional cationic poly-L-lysine layer on the bottom of each flowcell acts as a capture surface. When a voltage is applied, the negatively-charged bacterial/yeast cells migrate to the positively-charged capture surface where they are immobilized prior to imaging.
Cocktails of ATTO-532 (green) fluorescently-labelled DNA probes bind to the ribosomal RNA of target organisms following permeabilization. Each cocktail also includes ATTO-647 (red) labelled universal bacterial probe that binds to the ribosomal RNA of all clinically relevant bacteria (bacterial ID channels) or universal eukaryotic probe that binds to the ribosomal RNA of all clinically relevant yeast (yeast ID channels). The system images each flowcell using an epifluorescence microscope with camera at 532 nm, 647 nm and in dark-field. To exclude debris, only dark-field objects colocalized with universal probe signal are included in analysis. Colocalization of target probe signal and universal probe signal identifies a target organism.
Translation - Portuguese Após a preparação da amostra, as células recuperadas são pipetadas automaticamente em vários canais de células de fluxo da cassete de testes. Camadas condutoras de óxido de índio e estanho (ITO) transparente revestem as superfícies internas superior e inferior de cada canal da célula de fluxo e atuam como elétrodos. Uma camada catiónica de poli-L-lisina adicional no fundo de cada célula de fluxo atua como uma superfície de captação. Quando é aplicada tensão, as células bacterianas/leveduras com carga negativa migram para a superfície de captação com carga positiva, onde são imobilizadas antes da formação de imagens.
Misturas de sondas de ADN com marcador de fluorescência ATTO-532 (verde) ligam-se ao ARN ribossomal dos organismos visados após a permeabilização. Cada mistura inclui igualmente uma sonda bacteriana universal com marcador ATTO-647 (vermelho) que se liga ao ARN ribossomal de todas as bactérias clinicamente relevantes (canais de identificação [ID] bacteriana) ou sonda eucariótica universal que se liga ao ARN ribossomal de todas as leveduras clinicamente relevantes (canais de identificação [ID] de leveduras).
